Project-JRF Recruitment – Structural & Functional Analysis of FlhF

“Structural and Functional Analysis of FlhF: A Signal Recognition Particle (SRP) Type GTPase Involved in Flagellar Localization in Pseudomonas aeruginosa

This project aims to investigate FlhF, a highly conserved SRP-type GTPase that plays a crucial role in determining the correct number and spatial placement of flagella in Pseudomonas aeruginosa. FlhF is known to regulate flagellar biogenesis, ensuring that the flagellum is positioned at the appropriate cellular pole, which is essential for effective motility, host colonization, and pathogenicity.

The research involves understanding both the structural features and biochemical functions of FlhF. By examining its GTP-binding properties, interaction partners, and three-dimensional structure, the project aims to uncover how FlhF coordinates flagellar assembly at the molecular level.

This includes:

  • Determining high-resolution structural details using methods such as X-ray crystallography or biophysical techniques.
  • Performing protein purification, biochemical assays, and crystallization trials to characterize FlhF’s activity.
  • Exploring how structural variations influence its GTPase function and role in flagellar localization.

A deeper understanding of FlhF will provide insights into the mechanisms of bacterial motility, potentially revealing new antimicrobial targets or strategies to interfere with the motility-dependent virulence of Pseudomonas aeruginosa.
